生物活性：
PD 98059 is a potent, selective and reversible cell permeable inhibitor of MEK-1 (MAPKK1 or MAPKs ).
Acts by binding to the inactivated form of MEK, thereby preventing its phosphorylation by cRAF or MEK
kinase (IC50 = 2-7 μM). It has been shown to enhance embryonic stem cell self-renewal and has been
used to unravel the role of the MAPK cascade in biological systems. It Inhibits cell growth and proliferation
in acute myelogenous leukemia (AML) cell lines; causes G1 arrest by blocking p53-dependent p21
induction. PD 98059 has also been shown to enhance insulin-mediated increase in glutathione levels as
well as, independent of MEK1 inhibition, elevate primary cultured rat hepatocyte glutathione levels.
PD98059 is soluble in DMSO at 10-100 mM. For use with in vitro or cell-based assays, it may be diluted into
aqueous buffers to yield desired concentrations (<100 µM) with final DMSO concentrations of ~0.1%. IC50
values for inhibitory activity against MEK1 are around 5-10 µM. For experiments with cultured cells, we
recommends pretreating with this inhibitor for one hour prior to stimulation.
